Efek Kombinasi Ekstrak Daun Kumis Kucing (Orthosipon aristatus) Dan Daun Sirsak (Annona muricata L.) Terhadap Penurunan Kadar Gula Darah Mencit (Mus musculus)

Abstract


The Effect Of Combination Of Cat Wrish (Orthosipon aristatus) and Soursop (Annona muricata L.) Leave Extract On Decreasing The Blood Sugar Levels Of Mice (Mus musculus)

Orthosiphon aristatus and Annona muricate L are traditional medicines used in the treatment of diabetes in Indonesia. Orthosiphon aristatus in the treatment of diabetes and its complications mainly include inhibiting the activity of -amylase and -glucosidase, antioxidant and anti-inflammatory activity, regulating lipid metabolism, increasing insulin secretion, improving insulin resistance, increasing glucose absorption, promoting glycolysis, inhibiting gluconeogenesis, promoting glucagon secretion. -likepeptide-1 (GLP-1) and antiglycation activity. Phenolic acids, flavonoids and triterpenoids may be the main components for the hypoglycemic effect on O. aristatus. The purpose of this study was to determine the effect of the combination of cat whiskers leaf extract (EDK) and soursop leaf extract (EDS) on reducing blood sugar levels in mice (Mus musculus). This study is an experimental study with a randomized control trial pre and post test design using the glucose tolerance method. The test animals used were 15 male mice (Mus musculus) which were divided into 5 groups, namely group 1 control with 1% sodium carboxy methyl salicylic suspension, group 2, comparison group with 0.0065% glibenclamide suspension, group 3 combination EDK 3.5% (0.35 g/kgBW) and EDS 1.05% (105 mg/kgBW), group 4 combined EDK 7% (0.7 g/kgBW) and EDS 0.7% (70 mg/kgBW ), and group 5 a combination of 10.5% EDK (1.05 g/kgBW) and 0.35% EDS (35 mg/kgBW g) which was administered orally with a volume of 0.2 ml/20gBW in mice. The results showed that all treatment groups experienced a decrease in blood sugar levels at 120 minutes but only the combination group of 0.7% EDS and 7% EDK had a greater decrease than Na CMC and was comparable to glibenclamide.

Orthosiphon aristatus  dan Annona muricate L adalah obat tradisional yang digunakan dalam pengobatan diabetes di Indonesia. Orthosiphon aristatus dalam pengobatan diabetes dan fungsinya komplikasi terutama termasuk menghambat aktivitas -amilase dan -glukosidase, antioksidan dan aktivitas anti-inflamasi, mengatur metabolisme lipid, meningkatkan sekresi insulin, memperbaiki resistensi insulin, meningkatkan penyerapan glukosa, mempromosikan glikolisis, menghambat glukoneogenesis, mempromosikan sekresi glukagon-likepeptide-1 (GLP-1) dan aktivitas antiglikasi. asam fenolat, flavonoid dan triterpenoid mungkin menjadi komponen utama untuk efek hipoglikemia pada O. aristatus. Adapun tujuan penelitian ini adalah mengetahui efek kombinasi ekstrak daun kumis kucing (EDK) dan Ekstrak daun sirsak (EDS) terhadap penurunan kadar gula darah mencit (Mus musculus). Penelitian ini merupakan penelitian eksperimen dengan desain randomize control trial pre and post test dengan metode toleransi glukosa. Hewan uji yang digunakan adalah mencit jantan (Mus musculus) sebanyak 15 ekor yang dibagi menjadi 5 kelompok, yaitu kelompok 1 kontrol dengan suspensi Natrium Carboksimetilselulosa 1%,kelompok 2 yaitu kelompok pembanding dengan suspensi glibenklamid 0,0065%, kelompok 3 kombinasi EDK  3,5 % (0,35 g/kgBB) dan EDS 1,05 % (105 mg/kgBB), kelompok 4 kombinasi EDK 7 % (0,7 g/kgBB) dan EDS 0,7 % (70 mg/kgBB) , dan kelompok 5 kombinasi EDK 10,5 % (1,05 g/kgBB) dan EDS 0,35 % (35 mg/kgBB g) yang dimasukkan melalui per oral dengan volume 0,2 ml/20gBB mencit. Hasil penelitian menunjukkan seluruh kelompok perlakuan mengalami penurunan kadar gula darah pada menit ke 120 namun hanya kelompok kombinasi EDS 0,7% dan EDK 7% yang memiliki penurunan lebih besar dari Na CMC dan sebanding dengan glibenklamid.
