Isolasi, Identifikasi dan Aktivitas Antibakteri dari Fungi Endofit Daun Miana Terhadap Escherichia Coli dan Vibrio Cholerae

Abstract


Isolation, Identification, And Antibacterial Activity Of Miana Leaf Endophite Fungs Against Escherichia Coli And Vibrio Cholerae

Miana leaves have been proven empirically and scientifically to have the potential as an antibacterial that causes infections of the gastrointestinal tract, respiratory tract and skin. Miana comes from the Tana Toraja Regency and her cultivation requires sufficient land.  The antibacterial potential of miana leaves needs to be explored from the endophytic microbes it contains. The ability of endophytic microbes to produce secondary metabolites that are the same or similar to their host plants is a great opportunity to be exploited. Endophytic fungi are able to produce compounds that have potential as antioxidants, anticancer, antibacterial, antiviral, antifungal. Method. A step-by-step study of material preparation, culturing endophytic fungi on SDA media containing chloramphenicol. Isolation and purification of endophytic fungi isolates by repeated culturing, macroscopic and microscopic identification of the type of endophytic fungi isolates obtained, testing of the antibacterial activity of endophytic fungi isolates using the agar diffusion method. The results of the study found 4 isolates of endophytic fungi and all had potential as antibacterial. Conclusion. Fungal isolates found were isolates of white (Mucor sp), black (Aspergillus niger), green (Aspergillus fumigates) and gray (Rhizopus sp). All isolates can inhibit the growth of E.coli and Vibrio cholerae.

Daun miana telah terbukti secara empiris dan ilmiah berpotensi sebagai antibakteri  penyebab infeksi saluran cerna, salauaran napas dan kulit. Miana berasal dari Kabupaten Tana Toraja dan budidayanya membutuhkan lahan yang cukup. Potensi antibakteri daun miana perlu dieksplorasi dari mikroba endofit yang dikandungnya. Kemampuan mikroba endofit untuk menghasilkan metabolit sekunder yang sama atau mirip dengan tanaman inangnya merupakan peluang besar untuk dimanfaatkan. Fungi endofit mampu menghasilkan senyawa  yang berpotensi sebagai antioksidan, antikanker, antibakteri, antivirus, antifungi. Metode. Penelitian bertahap dari penyiapan bahan, mengkultur fungi endofit pada media SDA yang mengandung kloramfenikol. Isolasi dan pemurnian isolate fungi endofit dengan cara mengkultur berulang, identifikasi makroskopik dan mikroskopik jenis isolate fungi endofit yang diperoleh, pengujian  aktivitas antibakteri isolate fungi endofit dengan metode difusi agar. Hasil penelitian ditemukan 4 isolat fungi endofit dan semua berpotensi sebagai antibakteri. Kesimpulan. Isolate fungi yang ditemukan adalah isolate putih (Mucor sp), hitam (Aspergillus niger), hijau (Aspergillus fumigates) dan abu-abu (Rhizopus sp). Semua isolate dapat menghambat pertumbuhan E.coli dan Vibrio cholerae.
